Background:
-Amyloid is an extracellular filamentous protein
deposit found in the brain and is derived from
the amyloid beta-precursor protein. It is the
major protein component of amyloid cores,
neuritic plaques and is also found as a deposit in
neurfibrillary tangles. In humans, Alzheimer's
disease is the most common cause of senile
dementia and is characterized by abnormal
filamentous protein deposits in the brain. -
amyloid deposits are also detected in Lewy body
dementia, Down's syndrome, amyloidosis (Dutch
type and is Guam Parkinson-Dementia complex.
Specificity:
Reacts with Beta-Amyloid. It reacts with a 4-5
kDa peptide (termed amyloid beta protein/
amyloid A4 or amyloid beta peptide) derived
from the amyloid beta precursor protein. This
antibody detects extracellular amyloid beta
protein within senile plaques and vessel amyloid
in formalin-fixed, paraffin-embedded brain of
Alzheimer's and Lewy body disease.
Molecular Weight of Antigen:
102 kDa (precursor 4-5 kDa (mature).
Cellular Localization:
Transmembrane (APP) and extracellular
(amyloid ).
Species Reactivity:
Human, others not tested.
Ig Isotype:
IgG1/k
Immunogen:
A synthetic peptide consisting of residues 8-17
(SGYEVHHQKL) of the amyloid beta-protein,
which is the same as aa 660-669 of human
amyloid A4 / amyloid protein precursor.
Hybridoma:
Produced by the fusion between BALB/C
splenocytes and mouse myeloma p3-NS-1/Ag4-
1 (NS1) cells.
Format:
500 μL monoclonal antibody in tissue culture
supernatant with 0.09% sodium azide.
Positive Control:
Human Brain of Alzheimer's disease.
Storage:
Store at 4°C.
Applications and Suggested Dilutions:
Immunohistochemistry: Formalin/Paraffin only.
Use at a dilution of 1:25 to 1:50 for 60 min. at
RT. Staining of formalin-fixed tissues will be
enhanced by pretreatment with 95% formic
acid for 5-30 min at RT. Thorough washing
must be performed after the acid treatment.
The optimal dilution for a specific application
should be determined by the researcher.
